Generation of a phage display library for antibody selection and production. Human B lymphocytes are the source of a repertoire of antibody heavy and light chain gene segments (VH and VL) that are inserted in a pair-wise, random manner into a bacteriophage genome. Mature phage expresses the VH–VL polypeptides fused to phage coat proteins. Phages that express the desired VH–VL polypeptide can be selected and the corresponding antibody genes inserted into a cell line for expression and production
of antibody.
