Strontium ranelate activates components of canonical and noncanonical Wnt signaling in osteoblasts. Strontium ranelate activates the calcineurin/NFAT c1 signaling pathway in osteoblasts. This results in increased expression
of Wnt proteins (Wnt3a and Wnt5a). Wnt3a interacts with LRP5 (or LRP6) and Frizzled (Frz) coreceptors to activate canonical
Wnt/β-catenin signaling. Wnt5a interacts with the Ryk coreceptor to induce noncanonical RhoA-dependent signaling. These two
pathways converge to promote osteoblast replication and function.
